A Feedback Mechanism to Control Apoptosis Occurs in the Digestive Gland of the Oyster Crassostrea gigas Exposed to the Paralytic Shellfish Toxins Producer Alexandrium catenella ArchiMer
Rolland, Jean-luc; Medhioub, Walid; Vergnes, Agnes; Abi-khalil, Celina; Savar, Veronique; Abadie, Eric; Masseret, Estelle; Amzil, Zouher; Laabir, Mohamed.
To better understand the effect of Paralytic Shellfish Toxins (PSTs) accumulation in the digestive gland of the Pacific oyster, Crassostrea gigas, we experimentally exposed individual oysters for 48 h to a PSTs producer, the dinoflagellate Alexandrium catenella. In comparison to the effect of the non-toxic Alexandrium tamarense, on the eight apoptotic related genes tested, Bax and BI.1 were significantly upregulated in oysters exposed 48 h to A. catenella. Among the five detoxification related genes tested, the expression of cytochrome P450 (CYP1A) was shown to be correlated with toxin concentration in the digestive gland of oysters exposed to the toxic dinoflagellate. Beside this, we observed a significant increase in ROS production, a decrease in...

Azaspiracid Toxins: Toxicological Profile ArchiMer
Hess, Philipp; Twiner, Michael J; Kilcoyne, Jane; Sosa, Sylvio.
Azaspiracids (AZAs) are a toxin group that originate from marine dinoflagellates of the genera Azadinium and Amphidoma. After accumulation of these toxins in edible marine organisms and their subsequent consumption, humans develop a gastrointestinal syndrome referred to as azaspiracid shellfish poisoning (AZP). This syndrome is very similar to diarrheic shellfish poisoning (DSP), with main symptoms appearing after a few hours from consumption and including diarrhea, vomiting, and stomach cramps. Due to extensive metabolism in shellfish, more than 30 analogues have been reported to date, and purified compounds for selected analogues have recently been made available for toxicological studies. Currently, only AZA1, AZA2, and AZA3 are regulated in Europe and...

The REPAMO network (network ofmollusc pathology), created in 1986, ensures the survey of shellfish health status along French coasts according? to European Directives 91/67/EEC and 95/70/EEC. The aims of the network are 1 - to prevent the introduction and spread of exotic pathogens, 2-to study the means to decrease the impact of pathogens already present and to survey their evolution. In 2003, the REPAMO network focused on the survey oflisted diseases (bonamiosis and marteiliosis) in two areas under agreement process (Banc de Granville and zone X) and on the study of abnormal morlalities. Number of reported abnormal morlalities has increased in 2003. This rise can be partly explained by the recent network restructuring, but also by the summer heatwave and...

Biodiversity–Ecosystem Functioning (BEF) approach to further understanding aquaculture–environment interactions with application to bivalve culture and benthic ecosystems ArchiMer
Lacoste, Elise; Mckindsey, Christopher W.; Archambault, Philippe.
Coastal benthic ecosystems may be impacted by numerous human activities, including aquaculture, which continues to expand rapidly. Indeed, today aquaculture worldwide provides more biomass for human consumption than do wild fisheries. This rapid development raises questions about the interactions the practice has with the surrounding environment. In order to design strategies of sustainable ecosystem exploitation and marine spatial planning, a better understanding of coastal ecosystem functioning is needed so that tools to quantify impacts of human activities, including aquaculture, may be developed. To achieve this goal, some possible directions proposed are integrated studies leading to new concepts, model development based on these concepts and...

Biologie, abondance et cartographie de deux espèces de bivalves : l’huitre perlière <Pinctada radiata> et la coque glauque <Cerastoderma glaucum> dans le golfe de Gabes OceanDocs
Derbali, Abdelkarim.
The pearl oyster Pinctada radiata and the cockle Cerastoderma glaucum are among the most abundant bivalve molluscs in southern Tunisian waters. These species are not currently subject to any investigation despite their important economic value in the world. For this reason, their biological parameters, their spatial distribution and their stock assessment were undertaken in the Gulf of Gabes. The reproductive cycle was studied in relation to variations of the environment parameters. The specimens were collected monthly during one year (2007) in sites of El Kraten and El Jorf (Kerkennah Islands) for the first species and sites of Sidi Mansour and El Hicha (North of Sfax and Gabes) for the second. An intraspecific polymorphism biometric trait for P. radiata...

Calcein and manganese experiments for marking the shell of the common cockle (Cerastoderma edule): tidal rhythm validation of increments formation ArchiMer
Mahe, Kelig; Bellamy, Elise; Lartaud, Franck; De Rafelis, Marc.
This work focuses on investigating the potential of calcein and manganese as growth markers of the common cockle (Cerastoderma edule) in the bay of Somme (France). Recapture of shells, previously marked using a chemical marking and then bred in natural conditions, was performed in order to determine the shell growth patterns. Calcein marking has shown a fluorescent increment in shells after only 30 min immersion time at 150 mg L-1, but also for shells immersed 3 h at 50 mg L-1. Likewise, manganese shell marking was revealed under cathodoluminescence for shells immersed 1 h at 120 mg L-1 as well as for shells which spent 4 h at 90 mg L-1. A numerical analysis performed on each marked cockles has revealed 23 micro-increments between the mark and the ventral...

Cold storage of Pacific oysters out of water: biometry, intervalval water and sensory assessment ArchiMer
Buzin, Florence; Baudon, Violaine; Cardinal, Mireille; Barille, Laurent; Haure, Joel.
Due to the important economic activity of oyster production in France, microalgal toxic events represent a significant constraint for shellfish farmers who face closures of bivalve production-sites. The frequency of closures of 3 weeks or less represents half of the total closures along the French coasts. Cold storage could be a simple and affordable solution for temporary and short-term storage before commercialisation. A stock of marketable Pacific oyster Crassostrea gigas was therefore stored during 22 days in non-immersed conditions at 3 degrees C with 100% humidity. At the end of the experiment, the oyster mortality rate remained at a low level of less than 3.5%. The sensory attributes, odour, appearance, texture and flavour, did not show significant...

Comment améliorer la précocité de l'alerte en conchyliculture ? Exemple des mortalités de moules en 2014 dans les Pertuis Charentais ArchiMer
Lupo, Coralie; Prou, Jean.
Surveillance of shellf ish health is based on clinical surveillance of shellfish mortality, including a detection phase and a mandatory notification phase. While control measures are limited, early detection of pathogens is a challenge which needs to be met. The relevance and feasibility of an enhanced clinical surveillance based on phone interviews were studied. The results made it possible to reconstruct the spatial and temporal evolution of the outbreaks of mussel mortality which occurred in Pertuis Charentais in 2014. Moreover, a qualitative analysis of the interviews revealed contextual elements that were used to enhance the previous description and to devise causal scenarios which could be used as research hypotheses. By incorporating the...

Contamination of Clams with Human Norovirus and a Novel Hepatitis A Virus in Cameroon ArchiMer
Bonny, Patrice; Desdouits, Marion; Schaeffer, Julien; Garry, Pascal; Essia Ngang, Jean Justin; Le Guyader, Soizick.
Shellfish constitute an important protein source but may be contaminated by viruses from various origins. A study performed on clams collected in Cameroon showed a high prevalence of norovirus and hepatitis A virus. After sequencing, the hepatitis A virus showed similarities with the genotype V simian strains. The need for a one-health approach to protect humans from emerging diseases is evident by the high number of microbial pathogens that become zoonotic following insect bites or consumption of contaminated meat. This is particularly well documented for infectious disease transmission from non-human primates to human (Devaux et al. 2019). Environmental issues are critical in such events, especially for RNA viruses that may be excreted at high...

Cytotoxicity and mycotoxin production of shellfish-derived Penicillium spp., a risk for shellfish consumers ArchiMer
Geiger, Marie; Guitton, Yann; Vansteelandt, M.; Kerzaon, I.; Blanchet, Estelle; Du Pont, T. Robiou; Frisvad, J. C.; Hess, Philipp; Pouchus, Y. F.; Grovel, Olivier.
In order to assess the putative toxigenic risk associated with the presence of fungal strains in shellfish-farming areas, Penicillium strains were isolated from bivalve molluscs and from the surrounding environment, and the influence of the sample origin on the cytotoxicity of the extracts was evaluated. Extracts obtained from shellfish-derived Penicillia exhibited higher cytotoxicity than the others. Ten of these strains were grown on various media including a medium based on mussel extract (Mytilus edulis), mussel flesh-based medium (MES), to study the influence of the mussel flesh on the production of cytotoxic compounds. The MES host-derived medium was created substituting the yeast extract of YES medium by an aqueous extract of mussel tissues, with...

Défis de la surveillance des maladies chez les coquillages marins en France ArchiMer
Lupo, Coralie; Francois, Cyrille; Arzul, Isabelle; Garcia, Celine; Joly, Jean-pierre; Renault, Tristan.
The basic concepts and objectives of animal disease surveillance are common to all types of animal production. Yet, the procedures for implementation have been mostly developed for terrestrial animal productions. Disease surveillance in marine shellfish calls for specific measures in view of the peculiarities of marine biology and rearing techniques. The main objective of disease surveillance in marine shellfish is to detect as early as possible exotic and newly emerging diseases. With this objective in mind, a national surveillance network for surveillance of mollusc diseases called Repamo has been developed. It combines passive reporting and risk-based surveillance.

DEMAND INTERACTION BETWEEN FARMED SALMON AND WILD CAUGHT FISH IN THE UNITED KINGDOM AgEcon
Fofana, Abdulai; Clayton, Patty.
Demand relationships between salmon and a number of wild-caught whitefish and shellfish species using both single equation models and linearised AIDS system framework. The system is well represented although autocorrelation were found in both approaches but this is less of a problem in the systems approach. A cautious interpretation of the results indicated that salmon had a long-run market relationship with the whitefish species of cod, monkfish, saithe, whiting and plaice and with the shellfish species of mussels, nephrops, scallops and shrimp. These groups contain the main seafood species consumed within the United Kingdom, and therefore should include most potential substitutes for salmon.

Depuration: An administrative overview ArchiMer
Somerset, I.
At the 1964 National Shellfish Sanitation Workshop, Mr. Eugene Jensen presented a thought-provoking paper outlining the rationale for the depuration of shellfish. Mr. Jensen noted that "we are committed to the principal that shellfish must be as safe to eat as other ordinary food" and that "the problems of assuring the sanitary quality of coastal areas will increase rather than diminish. This is not the same as saying that pollution will increase" (Jensen, 1964). It is recognised that additional use of the waters and shore areas will have adverse impacts on the water quality and shellfish resource quantity and quality. If we are to maintain a program which will permit the consumer to eat shellfish in any form they choose, we must develop the means to...
